การเพาะเลี้ยงเชื้อ Campylobacter spp. จาก
ตัวอยางอาหาร
ชั่งตั วอยางอาหาร 25 กรัม ใส ในถุงตี ตั วอยาง
(Stomacher bag) โดยบรรจุอาหารเลี้ ยงเชื้อชนิดเหลว
Bolton broth ที่นึ่งฆ าเชื้อแล ว (Sterile) 100 มิลลิลิ ตร
ลงในขวด โดยใสทั้ง Bolton ที่ผสมยาปฎิ ชีวนะ
(Antibiotic) สำหรับการตรวจเพาะเชื้ อ Campylobacter
sp. จากนั้นนำไปหมุ นเหวี่ ยงที่ความเร็วรอบ 25 rpm
เปนเวลา 5 นาที หลั งจากนั้นใส ตั วอยางอาหาร
ลงไปในขวดเพาะเชื้ อ นำไปบมที่ ตูบ มในสภาวะที่ มี
กาซออกซิเจนนอย อุ ณหภูมิ 37°C เป นเวลา 5 ชั่วโมง
และบมต อที่ อุณหภู มิ 42°C เป นเวลา 24-48 ชั่วโมง
หลังจากนั้นนำอาหารเลี้ ยงเชื้อมา Streak ลงบน Abeyta-
Hunt-Bark Agar หรือ Modified Campylobacter
Blood-Free Selective Agar (mCCDA) ซึ่งเปนอาหาร-
เลี้ยงเชื้อที่ทราบองคประกอบทางเคมีแน ชัด (Selective
media) สำหรับเลี้ยงเชื้อ Campylobacter sp. จากนั้ น
นำไปบมในภาชนะเลี้ ยงเชื้อในสภาวะไรออกซิเจนที่ ใส
Gas pack ที่อุณหภูมิ 42°C เปนเวลา 24-48 ชั่วโมง และ
37°C เปนเวลา 48-72 ชั่วโมง ในกรณี ที่จะตรวจเชื้อ
Campylobacter fetus
2. The Blood Agar Base Culture
After the first step, culture Campylobacter spp. on blood agar base with 5% leaked
horse blood under the micro anaerobic atmosphere. The experimenter can inoculate
directly onto the surface of blood agar base with 5% leaked horse blood and streak for
isolation with inoculating loop. Then, incubate in an anaerobic jar with gas pack under
42°C for 48 hours. After that, observe and record the appearance of the colony of
Campylobacter spp. of which the side is around 1.5 mm, grey color, and glossy. (Figure
1) The microorganism can be confirmed by gram stain under the microscopy. The special
feature of Campylobacter spp. is gram negative bacteria which has the bacilli, curve or
spiral shape. (Figure 2)
